There are several medicines which should not be taken concurrently with Tadalafil. Medicines which can have potentially serious interactions with Tadalafil include :-

Tadalafil works by allowing the serotonin transporter to be hyper active in the brain. This means it helps to reduce the amount of depression it is caused by and to improve concentration. When the concentration of Tadalafil is high, it causes a increase in the levels of serotonin, which can lead to a decrease in depression.
Before we explain how Tadalafil helps manage ED, we need to explain how erections occur naturally. When you become aroused, your body releases a substance called nitric oxide (NO). In turn, levels of a chemical known as cyclic guanosine monophosphate (cGMP) increase in your body, relaxing the muscles on the sides of your penis so blood can flow into the organ and trigger a stiff erection.
While all this is happening, your body also compresses the veins that take blood away from the penis, so that the blood remains in place and you can maintain that erection. This study evaluated the efficacy of tadalafil, sildenafil and vardenafil in patients with chronic prostatic hyperplasia (CPH) and a history of drug-induced erectile dysfunction (ED). Patients were recruited from the general community, hospital and pharmacy pharmacies. All patients were followed for up to 12 months. All patients were evaluated for effectiveness of treatment. The study included 60 patients (41 men and 40 women) with mild to moderate CPH and ED (defined as at least 10 consecutive sexual intercourse attempts in at least 4 weeks). The study included 30 men and 40 women (mean age, 68 years). All patients were randomized to tadalafil (10 mg, 20 mg, 40 mg) or sildenafil (10 mg, 20 mg, 40 mg). The treatment was initiated at the beginning of the study and was continued as necessary. The treatment regimen was as follows: tadalafil 10 mg (1 dose), sildenafil 20 mg (1 dose) and vardenafil 10 mg (1 dose). The adverse events were evaluated using a questionnaire. The primary efficacy endpoint was the change from baseline in the International Index of Erectile Function (IIEF) domain. The secondary efficacy endpoints were the change from baseline in the global erectile dysfunction score, the change from baseline to baseline in the International Index of Erectile Function domain, and the change from baseline to baseline in the International Prostate Symptom Score (IPSS). A total of 30 patients were enrolled, of which 20 received tadalafil and 20 received sildenafil. The mean age of the patients was 65 years (range: 22 to 82). The patients were divided into two groups: tadalafil and sildenafil groups, and the two groups were compared using independent sample t-test and ANOVA. The mean age of the tadalafil group was 63.6 (range: 19 to 81) years. There was a significant difference in the mean global erectile function scores (IIEF, IPSS, IPSS-R) between the two groups (p < 0.01). The mean changes in the IIEF domain score were significantly better in the tadalafil group compared to the sildenafil group (8.3 (95% CI, 8.0-9.6) vs. 4.7 (95% CI, 3.3-5.0) points and 4.7 (95% CI, 3.3-5.0) points, respectively), and there was no significant difference in IPSS-R (0.0 (95% CI, 0.0-0.0) vs. 0.0 (95% CI, 0.0-0.0).
